Overview

BOXR1030 is a next-generation chimeric antigen receptor (CAR) T cell therapy designed to target glypican-3 (GPC3)-expressing solid tumors. It uniquely co-expresses a GPC3-targeted CAR and the exogenous glutamic-oxaloacetic transaminase 2 (GOT2) enzyme, which enhances T cell metabolism and function within the immunosuppressive tumor microenvironment. Preclinical studies have shown that BOXR1030 T cells exhibit improved proliferation, resistance to exhaustion, enhanced cytokine production profiles, and superior antitumor activity compared to standard CAR T cells in models of hepatocellular carcinoma, squamous cell carcinoma of the lung, myxoid/round cell liposarcoma, and Merkel cell carcinoma. The drug is being developed by SOTIO for use in patients with advanced GPC3-positive solid tumors[1][5][6][8].
