**BP-PEG-RC48-R848** is an experimental HER2-targeted black-phosphorus nanocomposite developed for photothermal-immunotherapy of HER2-positive breast cancer. It incorporates PEGylated two-dimensional black phosphorus as a near-infrared photothermal nanocarrier, the HER2-directed antibody-drug conjugate disitamab vedotin, and resiquimod, a Toll-like receptor 7 and Toll-like receptor 8 agonist. Disitamab vedotin directs the construct to HER2-expressing tumor cells and delivers the microtubule-disrupting payload monomethyl auristatin E, while resiquimod promotes dendritic-cell maturation and pro-inflammatory immune activation. In preclinical tumor models, near-infrared irradiation of the tumor-localized nanocomposite produced photothermal tumor ablation and enhanced antitumor immune responses.
