BPI-27336

BPI-27336 is a potent, selective, and orally bioavailable small molecule inhibitor of Extracellular Signal-Regulated Kinase 1 and 2 (ERK1/2), developed by Beta Pharma. It targets the terminal kinases of the Mitogen-Activated Protein Kinase (MAPK) signaling pathway, which is a critical driver of cell proliferation, differentiation, and survival. Dysregulation of this pathway, often through mutations in KRAS, NRAS, or BRAF, is a hallmark of many solid tumors. By inhibiting ERK1/2, BPI-27336 effectively blocks the signaling cascade regardless of upstream mutations, potentially overcoming resistance to BRAF or MEK inhibitors. The drug is currently undergoing Phase 1 clinical evaluation for the treatment of advanced solid tumors harboring MAPK pathway alterations.
