BPI-43487 is an orally bioavailable, highly selective small molecule inhibitor of Fibroblast Growth Factor Receptor 4 (FGFR4), developed by Beta Pharma. It is designed to target the FGF19/FGFR4 signaling pathway, which is frequently dysregulated in certain cancers, particularly hepatocellular carcinoma (HCC). By binding to FGFR4, BPI-43487 inhibits the downstream signaling cascades that promote tumor cell proliferation and survival. It is currently being evaluated in Phase 1 clinical trials for patients with advanced solid tumors, with a focus on those harboring FGF19 amplification or FGFR4 overexpression, including hepatocellular carcinoma and cholangiocarcinoma.
