Overview

BPI-501836 is a highly potent and selective small molecule inhibitor designed to target the KRAS G12D mutation. Developed by Betta Pharmaceuticals, the compound works by disrupting the interaction between the mutant KRAS G12D protein and its effector cRAF, thereby blocking the downstream MAPK signaling pathway and inhibiting tumor cell proliferation. In preclinical studies, BPI-501836 demonstrated significant anti-tumor efficacy in human pancreatic cancer xenograft models, showing tumor regression and sustained suppression of ERK phosphorylation. Its favorable pharmacokinetic profile, characterized by a prolonged half-life and high tumor distribution, supports a potential once-weekly dosing schedule. As of early 2024, BPI-501836 is in the IND-enabling stage of development.
