BPI-9208 is an investigational bispecific monoclonal antibody developed by Beta Pharma that simultaneously targets Leukocyte Surface Antigen CD47 and Programmed Death-Ligand 1 (PD-L1). CD47 is a transmembrane protein that acts as a "don't eat me" signal, inhibiting macrophage phagocytosis by binding to SIRPα; blocking this interaction enhances the innate immune system's ability to clear tumor cells. PD-L1 is a checkpoint protein that suppresses T-cell activity; its inhibition restores the adaptive immune response against cancer. By co-targeting these pathways, BPI-9208 is designed to provide synergistic anti-tumor activity. The bispecific format may also improve the safety profile by preferentially localizing CD47 blockade to PD-L1-expressing tumor tissues, potentially reducing the hematologic toxicities, such as anemia, often associated with systemic CD47 inhibition.
