BpMPLA is an immunological adjuvant developed by the Butantan Foundation, derived from the monophosphoryl lipid A (MPLA) of *Bordetella pertussis*. It functions as a potent agonist of Toll-like receptor 4 (TLR4), a key pattern recognition receptor on innate immune cells. By triggering TLR4 signaling, BpMPLA promotes the maturation of antigen-presenting cells and the secretion of pro-inflammatory cytokines, which in turn enhances the magnitude and quality of the adaptive immune response to co-administered vaccine antigens. Specifically, it is known to favor a Th1-biased immune response, making it particularly useful for vaccines targeting intracellular pathogens or those requiring strong cellular immunity. BpMPLA is part of Butantan's effort to develop proprietary adjuvant technologies for its vaccine pipeline, including potential applications in pertussis and influenza vaccines.
