Overview

**BPN14770 + donepezil** is an investigational combination regimen in which BPN14770, a first-in-class, selective small molecule inhibitor and negative allosteric modulator of phosphodiesterase-4D (PDE4D), is administered alongside donepezil, a widely used acetylcholinesterase inhibitor for Alzheimer’s disease. BPN14770 enhances synaptic plasticity and memory function by modulating the cAMP pathway in the brain, while donepezil increases cholinergic neurotransmission by inhibiting acetylcholinesterase. This combination is being studied to determine whether the synergistic targeting of both cyclic nucleotide and cholinergic pathways can more effectively treat cognitive impairment in Alzheimer’s disease and potentially other neurological disorders[1][2][3][4].
