Drug intelligence / Profile preview
BPR-608009 is a novel peptide-based theranostic candidate targeting Glypican-3 (GPC3) for the diagnosis and treatment of GPC3-positive hepatocellular carcinoma (HCC). Developed by Chengdu Brilliant Pharmaceutical using its proprietary BriPep™ technology, BPR-608009 is a cyclopeptide binder designed to achieve high tumor uptake and retention with rapid renal clearance, thereby improving the therapeutic window compared to existing GPC3-targeted radiopharmaceuticals. It can be complexed with various radioisotopes, including gallium-68 ($^{68}\text{Ga}$) for PET/CT imaging, and lutetium-177 ($^{177}\text{Lu}$) or actinium-225 ($^{225}\text{Ac}$) for targeted radionuclide therapy. Preclinical studies demonstrate high binding affinity ($K_D = 0.32\text{ nM}$) to human GPC3 and enhanced tumor growth inhibition in GPC3-positive HCC models. Early clinical evaluation includes a first-in-human PET/CT imaging study in patients with HCC.
