Overview

BPR001 is a first-in-class, orally administered small molecule drug candidate developed by BiPER Therapeutics for the treatment of gastrointestinal cancers, particularly gastric cancer. It acts as a selective inhibitor of BiP (also known as GRP78 or HSPA5), a key protein involved in cancer cell survival and resistance to therapy. By inhibiting BiP, BPR001 induces unresolved endoplasmic reticulum (ER) stress in tumor cells, leading to their death—a mechanism validated in both *in vitro* and *in vivo* studies. The drug has shown efficacy as a single agent and in combination with chemotherapies and immunotherapies. Its development targets patients whose tumors overexpress BiP, representing more than 50% of gastrointestinal cancer cases[1][3][6].
