BPT264 is a novel recombinant protein immunotherapy developed by Bright Peak Therapeutics for the treatment of autoimmune diseases. It is a chemically engineered interleukin-2 (IL-2) variant designed to selectively expand regulatory T cells (Tregs) while minimizing activation of other immune cell types. The molecule features enhanced binding to the IL-2 receptor alpha subunit (IL2Rα), ablated binding to the beta subunit (IL2Rβ), and weak interaction with the beta-gamma complex compared to wild-type IL-2. This "alpha-enhanced/beta-dead" profile enables potent and selective Treg expansion—demonstrated as over 30-fold in mice and up to 60-fold in non-human primates—with minimal effects on CD8+ T cells, NK cells, or eosinophils. The drug is PEGylated for half-life extension and improved pharmacokinetics. Preclinical studies show strong suppression of antigen-specific T cell-mediated inflammation in animal models[1][3][4][8].
