**BPT323** is a preclinical anti-tumor necrosis factor alpha interleukin-2 immunocytokine developed by Bright Peak Therapeutics for autoimmune diseases. It consists of the engineered regulatory T-cell selective interleukin-2 payload BPT263 site-specifically conjugated to the Fc region of the anti-tumor necrosis factor alpha monoclonal antibody adalimumab. BPT263 has enhanced affinity for interleukin-2 receptor alpha and lacks binding to interleukin-2 receptor beta, aiming to selectively activate and expand regulatory T cells while avoiding activation of cytotoxic CD8-positive T cells. The adalimumab component retains tumor necrosis factor alpha binding and blockade, potentially concentrating regulatory T-cell expansion in inflamed, tumor necrosis factor alpha-rich tissues.
