BPT543

BPT543 is a half-life extended, PEGylated interleukin-18 (IL-18) therapeutic developed by Bright Peak Therapeutics for the treatment of cancer. It is engineered to be resistant to the naturally occurring IL-18 binding protein (IL-18BP), which typically neutralizes IL-18 activity in the tumor microenvironment. BPT543 features an enhanced affinity for the IL-18 receptor and is created through site-specific conjugation of an optimized IL-18 payload to a 30 kDa PEG, significantly improving its pharmacokinetic and pharmacodynamic properties. Preclinical studies have shown that BPT543 exhibits potent antitumor activity, particularly when used in combination with PD-1 checkpoint inhibitors.
