Overview

BPX-101 is a first-generation, autologous dendritic cell-based cancer vaccine developed by Bellicum Pharmaceuticals for the treatment of metastatic castration-resistant prostate cancer (mCRPC). The vaccine is manufactured by transducing antigen-presenting cells with an adenoviral vector encoding an inducible human CD40 receptor (ih-CD40), followed by incubation with a protein containing the extracellular domain of prostate-specific membrane antigen (PSMA). Upon administration, patients receive intradermal injections of the vaccine, followed 24 hours later by intravenous infusion of rimiducid (AP1903), a small molecule dimerizer that activates ih-CD40. This activation provides temporally controlled, lymphoid-localized stimulation of dendritic cells to enhance immune responses against PSMA-expressing tumor cells. Clinical trials demonstrated that this approach induces robust PSMA-specific immunity and anti-tumor activity in mCRPC patients. Development for prostate cancer has been discontinued[1][2][4][5].
