Overview

BPX-201 is a second-generation autologous dendritic cell vaccine developed by Bellicum Pharmaceuticals for the treatment of prostate cancer. The therapy uses the patient’s own matured dendritic cells engineered with the proprietary DeCIDe “on switch” and trained to target prostate cancer cells expressing prostate specific membrane antigen (PSMA). These antigen-presenting cells are transduced with an adenoviral vector encoding inducible MyD88/CD40 signaling domains, which are then activated *in vivo* by administration of the dimerizer drug AP1903 (Rimiducid). This approach is designed to stimulate cytotoxic T lymphocyte responses against tumor cells. The vaccine was investigated primarily for metastatic castration-resistant prostate cancer (mCRPC), but development was discontinued after phase I trials[1][2][4][5][7].
