Overview

BPX-603 is an investigational autologous CAR-T cell therapy developed by Bellicum Pharmaceuticals for the treatment of solid tumors that overexpress or amplify human epidermal growth factor receptor 2 (HER2). It is the first dual-switch GoCAR-T product candidate from Bellicum and incorporates two key technologies: an inducible coactivation domain (MyD88/CD40, also called iMC) to enhance T-cell proliferation, persistence, and resistance to tumor microenvironment suppression; and CaspaCIDe safety switch technology for real-time control of efficacy and safety. The therapy is designed to improve upon previous HER2-targeted CAR-T approaches by enabling clinicians to activate or eliminate the infused T cells using small molecules. Clinical development focused on previously treated, locally advanced or metastatic HER2-positive solid tumors including breast cancer, gastrointestinal cancers (such as gastric cancer), ovarian cancer, endometrial cancer, and colorectal cancer. As of March 2023, clinical trials have been discontinued/suspended in these indications[1][3][4][5][6][7].
