Overview

BPX-701 is a T cell receptor (TCR)-engineered cell therapy designed to target malignant cells expressing the preferentially expressed antigen in melanoma (PRAME). It incorporates a natural high-affinity TCR specific for PRAME and is further engineered with an inducible caspase-9 (iCasp9) safety switch, allowing selective elimination of the modified T cells if needed. The therapy was developed as a controllable immunotherapy for cancers such as acute myeloid leukemia (AML), myelodysplastic syndromes (MDS), and uveal melanoma. Its mechanism of action involves immunologic cytotoxicity through recognition of PRAME on tumor cells by the engineered TCR, leading to targeted anti-tumor activity[1][2][4][6].
