BPX01

BPX01 is a topical gel formulation of the tetracycline-class antibiotic minocycline hydrochloride, developed to treat moderate-to-severe inflammatory nonnodular acne vulgaris by delivering fully solubilized minocycline directly to sebum-rich pilosebaceous units in the skin while minimizing systemic exposure.[1][3][7][9] As a semisynthetic tetracycline, minocycline inhibits bacterial protein synthesis by blocking aminoacyl-tRNA binding to the mRNA–ribosome complex, providing antibacterial activity against Cutibacterium (Propionibacterium) acnes and additional anti-inflammatory effects thought to contribute to acne lesion reduction.[1] BPX01 is a hydrophilic, non-oily, easy-to-use gel that achieves local skin concentrations of minocycline above the minimum inhibitory concentration for P. acnes with negligible plasma levels, aiming to preserve the efficacy profile of oral minocycline while reducing dose-dependent systemic adverse effects and the risk of antibiotic resistance.[1][3][10] In Phase II clinical trials, once-daily BPX01 (1% and 2%) demonstrated significant reductions in P. acnes colonization and inflammatory lesion counts, with good cutaneous tolerability and very low systemic minocycline exposure.[1][4][6]
