Drug intelligence / Profile preview
BR-11257 is a novel, monocarboxylic, heme-independent soluble guanylate cyclase (sGC) activator developed by Bayer in collaboration with the University of Braunschweig. Unlike traditional dicarboxylic sGC activators (such as cinaciguat or BAY 60-2770) which strongly thermostabilize sGC and can cause severe hypotension, BR-11257 acts as a non-stabilizing partial agonist of sGC. This unique mechanism of action allows for gentle sGC activation, potentially avoiding excessive blood pressure lowering while maintaining therapeutic efficacy. Preclinical studies have demonstrated that BR-11257 has renoprotective effects, significantly lowering blood pressure, ameliorating proteinuria, and reducing glomerulosclerosis in chronic kidney disease (CKD) models with volume/salt-dependent hypertension.
