Overview

BR111 is a first-in-class antibody-drug conjugate (ADC) developed by BioRay Pharmaceutical. It targets dual non-overlapping epitopes of the receptor tyrosine kinase ROR1, which is highly expressed in various hematological malignancies and solid tumors but minimally present in normal tissues. The ADC consists of a biparatopic monoclonal antibody conjugated via BioRay's proprietary CysX™ irreversible site-specific technology to the small-molecule toxin eribulin. This design enables strong binding and efficient internalization into ROR1-positive tumor cells, where the cytotoxic payload is released to induce cell death. Preclinical studies have shown that BR111 has superior anti-tumor efficacy and safety compared to existing clinical alternatives, with additional effects such as bystander killing and immune activation within the tumor microenvironment. It is being developed for cancers including lymphoma, breast cancer, ovarian cancer, and lung cancer[1][2][5].
