Bradanicline is a novel small molecule that acts as a highly selective agonist (partial or full, depending on the source) at the alpha7 subtype of neuronal nicotinic acetylcholine receptors (α7 nAChR). It was originally developed by Targacept and later studied in collaboration with AstraZeneca. Bradanicline was investigated for its potential to treat central nervous system disorders, particularly cognitive impairment associated with schizophrenia, Alzheimer's disease, and attention-deficit hyperactivity disorder (ADHD). The drug demonstrated cognitive-enhancing effects in animal models and showed neuroprotective properties by modulating inflammation and protecting neurons from degeneration. Despite reaching phase II clinical trials for several indications—including schizophrenia (cognitive/negative symptoms), Alzheimer's disease, ADHD, and refractory chronic cough—development has been discontinued for all major CNS indications due to lack of efficacy or strategic decisions. Bradanicline remains primarily a research tool for studying α7 nAChR function[1][2][3][4][5][6].
