Overview

Brain-derived neurotrophic factor plasmid DNA is a non-viral, naked plasmid DNA gene therapy developed by UNSW Sydney (Translational Neuroscience Facility) for the treatment of severe to profound sensorineural hearing loss and deafness. Delivered via close-field electroporation (gene electrotransfer) using a cochlear implant electrode array directly into the mesenchymal cells of the cochlea during surgery, the plasmid encodes both brain-derived neurotrophic factor (BDNF) and neurotrophin-3 (NT-3). Once transfected, these cells produce BDNF and NT-3, which act as agonists on the TrkB and TrkC receptors of cochlear spiral ganglion cells, stimulating the regrowth of spiral ganglion peripheral neurites toward the cochlear implant electrodes to improve the neural interface.
