Overview

Brazikumab is a fully human monoclonal antibody that selectively targets the p19 subunit of interleukin-23 (IL-23), a pro-inflammatory cytokine implicated in the pathogenesis of inflammatory bowel diseases such as Crohn’s disease and ulcerative colitis. By binding to IL-23 and preventing its interaction with its receptor, brazikumab inhibits downstream signaling involved in inflammation. The drug was developed for patients with moderate-to-severe active Crohn’s disease and ulcerative colitis who had failed or were intolerant to at least one anti-tumor necrosis factor alpha agent. Clinical trials demonstrated that brazikumab was well tolerated over long-term treatment periods; however, development for these indications was discontinued due to strategic considerations rather than safety concerns[2][3][4][6].
