BRB-002 is a novel, investigational protein therapy developed by Bitterroot Bio for the treatment of atherosclerotic cardiovascular disease (ASCVD). It functions as an immunomodulatory agent that inhibits the CD47 cell surface receptor—often referred to as the “don’t eat me” signal—which plays a key role in preventing macrophage-mediated clearance of diseased cells. By blocking CD47, BRB-002 aims to modulate immune responses within atherosclerotic plaques, reducing vascular inflammation and plaque burden. This mechanism is distinct from current standards of care and targets underlying causes of ASCVD rather than just managing symptoms. Preclinical studies have shown that even partial occupancy of CD47 can meaningfully reduce plaque burden. The drug has demonstrated safety and robust target engagement in Phase 1 clinical trials with no serious adverse events reported[1][3][5][6][8].
