Overview

BRCAT431 shRNA is a short hairpin RNA (shRNA) therapeutic designed to target and knock down the long noncoding RNA (lncRNA) Breast Cancer Associated Transcript 431 (BRCAT431). Developed by researchers at the University of Michigan, BRCAT431 shRNA is being investigated for the treatment of estrogen receptor (ER)-positive breast cancer, particularly cases that have developed resistance to tamoxifen. BRCAT431 is an ER-regulated lncRNA that is overexpressed in breast cancer and interacts with the RNA-binding protein hnRNPL to promote aggressive oncogenic phenotypes, including cell proliferation, colony formation, and invasion. In preclinical studies, shRNA-mediated knockdown of BRCAT431 significantly reduced tumor growth and invasion in vitro and in vivo, and reversed tamoxifen resistance in breast cancer cell lines.
