Drug intelligence / Profile preview
BRD5529 is a small-molecule inhibitor of the caspase recruitment domain-containing protein 9 (CARD9) pathway. Developed by the Broad Institute, BRD5529 acts as a protein-protein interaction (PPI) inhibitor that disrupts the binding of CARD9 to the E3 ubiquitin ligase TRIM62 (tripartite motif-containing protein 62). By preventing TRIM62-mediated ubiquitination of CARD9, BRD5529 blocks downstream NF-κB activation and the subsequent release of proinflammatory cytokines. It has been investigated preclinically for the treatment of inflammatory bowel disease (IBD), axial spondyloarthritis (axSpA), and Pneumocystis pneumonia (PCP)-related lung injury.
