Drug intelligence / Profile preview
BRD7635 is a preclinical small molecule cargo receptor complex disruptor developed by the Broad Institute. It specifically binds to Transmembrane emp24 domain-containing protein 9 (TMED9) and disrupts the hetero-oligomeric TMED7/2/9/10 entrapment complex in the early secretory pathway. This complex is responsible for anchoring and sequestering misfolded mutant proteins, such as frameshifted mucin 1 (MUC1-fs), mutant semaphorin 4A (F350C), and mutant rhodopsin (P23H), within the Golgi apparatus. By disrupting this entrapment complex, BRD7635 promotes the lysosomal degradation of these toxic mutant proteins. It is currently in preclinical development for the treatment of proteinopathies, including Mucin 1 kidney disease (MKD), retinitis pigmentosa, and retinal detachment.
