BrE-3 ricin A-chain is an experimental **immunotoxin** created by the chemical conjugation of the monoclonal antibody BrE-3 (which targets the polypeptide core of the MUC1 mucin gene product) to the **A-chain of ricin**, a potent ribosome-inactivating protein. The immunotoxin is designed to selectively target and kill human small-cell lung cancer (SCLC) cells expressing MUC1 mucin by delivering ricin A-chain's catalytic activity directly to the tumor cell. The mechanism of cytotoxicity relies on ricin A-chain’s **rRNA N-glycosidase activity**, which inactivates ribosomes in the target cells, thereby irreversibly inhibiting cellular protein synthesis and inducing cell death. Preclinical studies demonstrated high potency and selectivity for SCLC cell lines in vitro.
