Brenetafusp (IMC-F106C) is a novel ImmTAC (Immune mobilizing monoclonal TCRs Against Cancer) bispecific fusion protein that targets PRAME (Preferentially Expressed Antigen in Melanoma) and CD3. It is designed to redirect and activate T cells against PRAME-expressing tumor cells. Pembrolizumab is a humanized monoclonal antibody that blocks the PD-1 receptor on T cells, preventing cancer cells from evading immune detection. The combination of brenetafusp and pembrolizumab aims to enhance anti-tumor immune responses by both directly targeting tumor antigens and releasing immune checkpoint inhibition. This combination has shown promising safety and efficacy in early-phase clinical trials for advanced or metastatic cutaneous melanoma patients who have been previously treated with immune checkpoint inhibitors[2][4][6][8].
