**Brenetafusp + tebentafusp** is a combination of two ImmTAC (immune mobilizing monoclonal TCRs against cancer) bispecific biologic drugs, both developed by Immunocore. Brenetafusp targets PRAME (preferentially expressed antigen in melanoma) and engages CD3 on T cells, while tebentafusp targets gp100 and also engages CD3. Both agents redirect cytotoxic T cells to tumor cells expressing their respective antigens through HLA-A*02:01 presentation. Tebentafusp is approved for unresectable or metastatic uveal melanoma and demonstrates overall survival benefit, while brenetafusp is in phase 1 studies primarily for ovarian cancer. Both drugs, when combined or with other agents, are under investigation for synergistic efficacy in enhancing antitumor immune responses in solid tumors resistant to other immunotherapies[3][4][1][2].
