Overview

This regimen is a **combination therapy** composed of brentuximab vedotin, rituximab, cyclophosphamide, doxorubicin, and prednisone. It is used for the treatment of B-cell lymphomas, particularly **CD30-positive lymphomas** such as diffuse large B-cell lymphoma (DLBCL), primary mediastinal B-cell lymphoma (PMBCL), and gray zone lymphoma (GZL). Brentuximab vedotin is an **antibody-drug conjugate** targeting CD30, delivering a cytotoxic antitubulin agent directly to tumor cells. Rituximab is a monoclonal antibody directed against CD20 on B-cells, inducing cell death via immune-mediated mechanisms. Cyclophosphamide and doxorubicin are small molecule **chemotherapeutic agents** with alkylating and DNA intercalating/topoisomerase II inhibitory action, respectively. Prednisone is a synthetic glucocorticoid providing anti-inflammatory and immunosuppressive effects. This combination is sometimes referred to as BV-R-CHP, substituting brentuximab vedotin for vincristine in the classic R-CHOP regimen. In clinical studies, this regimen has demonstrated high rates of complete response and promising survival outcomes for patients with CD30-positive B-cell lymphomas[2][7].
