Overview
A five-drug combination regimen consisting of **brentuximab vedotin** (an antibody-drug conjugate targeting CD30), **doxorubicin** (an anthracycline cytotoxic chemotherapeutic), **bleomycin** (a glycopeptide antineoplastic with DNA-damaging activity), **vinblastine** (a vinca alkaloid antimitotic), and **dacarbazine** (an alkylating agent). This experimental regimen is being studied as first-line therapy for classical Hodgkin lymphoma (cHL), aiming to improve therapeutic outcomes especially for advanced-stage or high-risk patients. **Brentuximab vedotin** delivers the cytotoxic payload MMAE to CD30-expressing Hodgkin Reed–Sternberg cells, while the other agents provide broad cytotoxic activity through various mechanisms. Early-phase clinical trials have investigated safety and feasibility, but there is currently no regulatory approval of this *five-drug* regimen for any indication[1].
