Overview

**Brentuximab vedotin + midazolam** is a combination of two drugs: brentuximab vedotin, a CD30-directed antibody-drug conjugate used as an anti-neoplastic agent in various lymphomas, and midazolam, a short-acting benzodiazepine used for sedation, anxiolysis, amnesia, and treatment of seizures. Brentuximab vedotin consists of an anti-CD30 monoclonal antibody linked to the microtubule-disrupting agent monomethyl auristatin E (MMAE); it targets and releases MMAE inside CD30-positive tumor cells, causing cytotoxicity. Midazolam acts as a positive allosteric modulator at the GABA-A receptor, enhancing inhibitory neurotransmission in the brain, which leads to sedation, anxiolysis, anticonvulsant, and amnestic effects. There is no unique clinical indication for the combination; instead, clinical studies show that brentuximab vedotin does not affect midazolam pharmacokinetics, and the combination may be of interest in studying drug-drug interactions during cancer treatment[3][5].
