brentuximab vedotin + nivolumab

Brentuximab vedotin + nivolumab is a combination regimen comprising brentuximab vedotin, an antibody-drug conjugate targeting CD30, and nivolumab, a monoclonal antibody checkpoint inhibitor against programmed death-1 (PD-1). Brentuximab vedotin delivers the cytotoxic agent monomethyl auristatin E to CD30-expressing tumor cells, leading to microtubule disruption and apoptosis. Nivolumab blocks PD-1, restoring T-cell-mediated anti-tumor immunity. This combination is being investigated primarily for relapsed or refractory classical Hodgkin lymphoma and primary mediastinal large B-cell lymphoma. Synergy is hypothesized via both immunogenic cell death and modulation of the tumor microenvironment, augmenting effects of immune checkpoint blockade[1][2][4][5][7].
