Bretovameran is the active ingredient in Comirnaty JN.1, an mRNA-based COVID-19 vaccine developed to target the Omicron JN.1 subvariant of SARS-CoV-2. It consists of a single-stranded, 5'-capped messenger RNA (mRNA) produced via cell-free in vitro transcription from DNA templates encoding the viral spike (S) protein of SARS-CoV-2 (Omicron JN.1). The mRNA is nucleoside-modified and embedded in lipid nanoparticles to facilitate delivery into host cells after intramuscular injection[4][7]. Once inside host cells, the mRNA is translated into the spike protein, which is then expressed on the cell surface and induces neutralizing antibody and cellular immune responses against SARS-CoV-2[6][8]. This mechanism aims to confer protection against COVID-19.
