Drug intelligence / Profile preview
Bria-IMT + immune checkpoint inhibitor is an investigational combination immunotherapy regimen being developed by BriaCell Therapeutics for the treatment of advanced or metastatic breast cancer. The regimen combines Bria-IMT (SV-BR-1-GM), an off-the-shelf, genetically engineered, irradiated allogeneic whole-cell breast cancer vaccine, with an immune checkpoint inhibitor (such as the anti-PD-1 monoclonal antibodies retifanlimab or pembrolizumab). Bria-IMT is engineered to secrete granulocyte-macrophage colony-stimulating factor (GM-CSF) and express HLA class I and II molecules, which directly and indirectly stimulate the patient's immune system to recognize and attack breast cancer cells. The addition of an immune checkpoint inhibitor further potentiates this anti-tumor activity by blocking the PD-1/PD-L1 pathway, thereby overcoming the immunosuppressive tumor microenvironment. The combination is currently being evaluated in a pivotal Phase III clinical trial (BRIA-ABC) for patients with heavily pretreated metastatic breast cancer.
