Bria-OTS is a personalized, "off-the-shelf" cellular immunotherapy developed by BriaCell Therapeutics for the treatment of advanced solid tumors, primarily metastatic breast cancer. As a next-generation version of the Bria-IMT platform, Bria-OTS consists of genetically engineered human breast cancer cell lines (such as the BC1 line) that are matched to a patient's HLA type. These cells are engineered to express exogenous MHC class II alleles and immune-stimulating cytokines, acting as a personalized tumor cell vaccine that activates the patient's immune system to recognize and destroy cancer cells. The therapy is typically administered as part of a regimen that includes low-dose cyclophosphamide and peginterferon alpha-2a. Bria-OTS is currently being evaluated in Phase 1/2 clinical trials both as a monotherapy and in combination with immune checkpoint inhibitors such as tislelizumab.
