Overview

Briciclib alaninate is a small molecule inhibitor of Cyclin D1 expression, originally developed by Onconova Therapeutics (as ON 013105) and licensed to Zai Lab for development in Greater China. It functions by inhibiting the translation of Cyclin D1 mRNA, likely through interaction with the eukaryotic translation initiation factor 4E (eIF4E) or the eIF4F complex, which is often overexpressed in various cancers. This downregulation leads to cell cycle arrest in the G1 phase and subsequent apoptosis. The drug has been primarily investigated for the treatment of advanced hepatocellular carcinoma (HCC) and other solid tumors. In a Phase 3 trial (CTR20180831) conducted by Zai Lab, it was evaluated as a second-line treatment for patients with advanced HCC.
