Drug intelligence / Profile preview
Bridging-BiTE (B-BiTE) is a novel bispecific T-cell engager (BiTE) modality developed by researchers at Ehime University for the treatment of multiple myeloma and other refractory hematological malignancies. B-BiTE is designed as a bispecific molecule containing two single-chain variable fragments (scFvs): one specific for the Fc domain of human immunoglobulin G (IgG) and the other specific for the CD3ε subunit of the T-cell receptor complex. When mixed with clinically available monoclonal antibodies (mAbs) such as daratumumab or elotuzumab, B-BiTE binds to the mAb's Fc region to form a mAb/B-BiTE complex. This complex redirects T-cell cytotoxicity against tumor cells via CD3ε binding while preserving and supporting natural killer (NK) cell reactivity through the mAb's Fc region, resulting in dual-lymphoid activation and enhanced anti-tumor efficacy.
