BRII-179 is a novel recombinant protein-based therapeutic vaccine (immunotherapeutic) designed to treat chronic hepatitis B infection. It is based on virus-like particles (VLPs) and expresses the Pre-S1, Pre-S2, and S envelope proteins of the hepatitis B virus (HBV), building upon the 3-antigen conformation of the prophylactic Sci-B-Vac vaccine. The mechanism of action involves priming both B-cell and T-cell immunity by inducing HBV-specific antibody and T-cell responses. This includes neutralizing circulating HBV, blocking infection of hepatocytes via Pre-S1-mediated mechanisms, and enabling immune-mediated clearance of infected hepatocytes. Clinical studies have shown that BRII-179 alone or in combination with pegylated interferon alpha can enhance HBV-specific immune responses in patients with chronic hepatitis B but has limited effect on virological status as monotherapy; its potential for achieving functional cure is being evaluated in combination regimens[1][4][6][8].
