Overview

BRII-5395 is an investigational therapeutic vaccine developed by Brii Biosciences for the treatment of advanced hepatitis B virus (HBV)-related hepatocellular carcinoma (HCC). It is a recombinant protein-based vaccine designed to stimulate a specific immune response against HBV antigens, including the surface antigen (HBsAg) and the Pre-S1 and Pre-S2 regions. The goal of this therapy is to overcome immune tolerance in chronic HBV patients and enhance T-cell activity to target and eliminate HBV-infected cells and associated tumor cells. In clinical development, BRII-5395 is being evaluated in combination with the PD-1 inhibitor sintilimab and the VEGF inhibitor bevacizumab to leverage potential synergies between therapeutic vaccination, immune checkpoint inhibition, and anti-angiogenic therapy in the HCC setting.
