BRII-732 is a medoxomil carbonate prodrug of islatravir (ISL/EFdA), a potent HIV-1 nucleotide reverse transcriptase translocation inhibitor. It is designed for once-weekly dosing as part of combination antiretroviral therapy for HIV. Following oral absorption, BRII-732 is efficiently converted to islatravir in vivo with no measurable systemic exposure to the prodrug itself. It demonstrates a long half-life, dose-dependent pharmacokinetic profile, and efficient intracellular formation of ISL-TP in peripheral blood mononuclear cells (PBMCs).
