BRII-753 is an investigational long-acting nucleoside reverse transcriptase translocation inhibitor (NRTTI) being developed by Brii Biosciences for the treatment and prevention of HIV-1 infection. As a prodrug of 4'-ethynyl-2-fluoro-2'-deoxyadenosine (EFdA, also known as islatravir), BRII-753 is designed as a subcutaneous injectable formulation with the potential for extended dosing intervals, including once-monthly, once-quarterly, or twice-yearly administration. The drug functions by inhibiting the HIV reverse transcriptase enzyme, specifically through a translocation inhibition mechanism that prevents the addition of subsequent nucleotides to the viral DNA chain. While it has completed Phase 1 clinical studies, Brii Biosciences announced in 2024 that it is seeking partnership opportunities for the asset to focus on its hepatitis B portfolio.
