Overview

Brimapitide (AM-111) is a synthetic, cell-permeable peptide inhibitor of c-Jun N-terminal kinase (JNK). Developed by Auris Medical (now Altamira Therapeutics), it was designed to protect the delicate sensory cells of the inner ear from stress-induced apoptosis following acute injury, such as sudden sensorineural hearing loss (SSNHL) or acoustic trauma. The drug is administered via a single intratympanic injection of a biocompatible gel, allowing for local delivery to the cochlea and sustained exposure. Despite showing promise in Phase 2 trials, the Phase 3 HEAL trial did not meet its primary efficacy endpoint in the overall study population, leading to the termination of its development for this indication.
