Brivanib + irinotecan is an investigational combination therapy composed of two small molecule drugs used primarily in the treatment of metastatic colorectal cancer and other gastrointestinal malignancies. Brivanib is an orally available prodrug that acts as a dual inhibitor of vascular endothelial growth factor receptor 2 (VEGFR-2) and fibroblast growth factor receptors 1 and 2 (FGFR-1/2), thereby blocking angiogenesis essential for tumor growth[5]. Irinotecan is a topoisomerase I inhibitor that interferes with DNA replication in cancer cells, leading to cell death. The combination aims to target both tumor vasculature (via antiangiogenic effects) and directly induce cytotoxicity in proliferating tumor cells. This regimen has been evaluated in phase I/II clinical trials for metastatic colorectal cancer, particularly after failure of standard therapies[1][7][8]. Brivanib was developed by Bristol Myers Squibb.
