Overview

Brivanib alaninate + doxorubicin is a combination therapy investigated for the treatment of advanced or metastatic solid tumors, including hepatocellular carcinoma. Brivanib alaninate (BMS-582664) is an oral prodrug of BMS-540215, a small molecule tyrosine kinase inhibitor that dual-targets vascular endothelial growth factor receptors (VEGFR) and fibroblast growth factor receptors (FGFR), which are key drivers of angiogenesis and tumor growth. Doxorubicin is a well-established anthracycline chemotherapy agent that works by intercalating into DNA and inhibiting the enzyme topoisomerase II, leading to DNA damage and cell death. The combination was evaluated in Phase I clinical trials by Bristol-Myers Squibb to assess safety, pharmacokinetics, and the maximum tolerated dose. However, development of brivanib alaninate was ultimately discontinued following disappointing results in Phase III trials for hepatocellular carcinoma as a monotherapy and in other combinations.
