Overview

Brivekimig is a **dual-target Nanobody (single-domain antibody) molecule** developed by Sanofi, designed to inhibit both **tumor necrosis factor alpha (TNF)** and **OX40 ligand (OX40L)**. It is being investigated primarily for the treatment of moderate to severe hidradenitis suppurativa (HS), but is also in phase II trials for Crohn’s disease, ulcerative colitis, focal segmental glomerulosclerosis, and type 1 diabetes mellitus. Brivekimig acts by reducing inflammation through simultaneous inhibition of two immune regulatory pathways, which is hypothesized to more effectively control inflammatory and immune-mediated disorders. In clinical studies, subcutaneous administration of brivekimig demonstrated clinically meaningful efficacy and was well tolerated, with nasopharyngitis and headache being the most frequent adverse events[1][2][3][4][5][6][7][8].
