Overview

Brivudine monophosphate is the phosphorylated derivative of brivudine, a nucleoside analog structurally related to thymidine. It belongs to the class of pyrimidine deoxyribonucleoside monophosphates and serves as an intermediate metabolite in the activation pathway of brivudine. Brivudine itself is an antiviral agent used primarily for herpes zoster (shingles) and acts by being converted intracellularly into its active triphosphate form via sequential phosphorylation steps. The triphosphate form is incorporated into viral DNA by viral DNA polymerase, leading to chain termination and inhibition of viral replication. Brivudine monophosphate represents the first phosphorylation product in this process but does not itself have direct antiviral activity; further phosphorylation to the triphosphate is required for efficacy[1][6][3]. Research also suggests that brivudine derivatives may downregulate DNA repair enzymes such as APEX nuclease 1 and inhibit STAT3 expression, potentially affecting apoptosis and chemoresistance pathways[6].
