BRL 42715

BRL 42715 is a novel penem compound that functions as a potent beta-lactamase inhibitor. It was developed by Beecham Pharmaceutical Research Division (later SmithKline Beecham Pharmaceuticals). The drug is designed to inhibit a wide range of bacterial beta-lactamases, including both plasmid-mediated and chromosomally-mediated enzymes, thereby restoring the efficacy of beta-lactam antibiotics like amoxicillin against resistant bacterial strains. In vitro studies demonstrated its ability to render various resistant bacteria, such as Klebsiella, Enterobacter, Citrobacter, Morganella, Serratia, Acinetobacter, Aeromonas, and Staphylococcus aureus, susceptible to amoxicillin. However, BRL 42715 was found to be too unstable for further development.
