Overview

BRL-50481 is a potent and highly selective small molecule inhibitor of phosphodiesterase 7 (PDE7), with significant activity against both PDE7A and PDE7B isoforms. By specifically blocking the hydrolysis of cyclic adenosine monophosphate (cAMP), BRL-50481 increases intracellular cAMP levels, which in turn modulates various downstream signaling pathways including the cAMP/PKA/CREB and AKT/GSK3β axes. Preclinical research has demonstrated its potential therapeutic utility across a broad range of conditions, including substance use disorders (specifically morphine addiction), inflammatory lung diseases like asthma and idiopathic pulmonary fibrosis, neurological disorders such as anxiety and neurodegeneration, and certain cancers like chronic lymphocytic leukemia (CLL). It is widely utilized as a pharmacological tool to investigate the role of PDE7 in immune response, neuroplasticity, and cell differentiation.
